NOURO Preclinical Research: The AAIC 2026 Study

NOURO's formulation was tested in a preclinical study at the Duke Molecular Physiology Institute and presented at the Alzheimer's Association International Conference (AAIC) 2026. This page documents that study in full, including what it does and does not show. It is animal research. Nothing here has been established in humans, and NOURO is a dietary supplement, not a treatment for any disease.

The study

"Can a novel mixture of natural supplements (Nouro™) protect against neurodegeneration in an APOE animal model of Alzheimer's disease?"

Alzheimer's disease involves several interrelated systems at once: beta-amyloid production, inflammation, immune response, cellular degradation, energy production and neuronal plasticity. The hypothesis behind this study was that no single-target compound would address that, and that a combination acting on several pathways would be needed. The study tested whether the NOURO formulation could do so in an animal model.

Design and methods

Model APP/PS1/APOE3 (AE3) and APP/PS1/APOE4 (AE4) knock-in mice, selected as the best available animal model of APOE-related sporadic Alzheimer's disease
Design From two months of age, mice were maintained on a low-fat diet or a high-fat diet, with or without the formulation
Duration Six months, after which plasma and tissues were collected
Formulation in diet Nicotinic acid 2194 mg/kg, alpha-lipoic acid 656 mg/kg, Siliphos 1250 mg/kg, berberine 1101 mg/kg, taurine 1650 mg/kg. These are concentrations in mouse feed and are not human doses.

Findings

In the animals that received the formulation, the researchers reported:

  • Weight. Control mice on the high-fat diet gained substantial weight over the study. Mice receiving the formulation did not, despite eating the same amount of food.
  • BDNF increased. Brain-derived neurotrophic factor, a protein associated with neuronal growth and plasticity, was higher in treated animals.
  • Amyloid plaque buildup was reduced.
  • Hippocampal volume was preserved. Treated animals did not show the loss of hippocampal volume seen in controls.
  • Glial markers were lowered in both AE3 and AE4 animals, and soluble APOE3 levels increased.

How the ingredients are thought to work

The five ingredients in the tested formulation were selected for mechanisms that act together rather than on a single target:

  • AMPK activation
  • NAD⁺/NADP⁺ enhancement
  • mTOR signalling inhibition
  • Antioxidant defence
  • Anti-inflammatory signalling

The researchers suggest the observed changes are likely due to the formulation restoring metabolic and neuronal proteostasis, the balance of protein production and clearance inside cells.

How to read these results

Limitations, stated plainly.

  • This is an animal study. Results in mice frequently do not transfer to people, and none of these findings has been established in humans.
  • It is preclinical, meaning it sits at an early stage of evidence, well before human trials.
  • The study was funded by Tonum Health, the maker of NOURO.
  • The tested diet formulation is not the retail product. Doses are feed concentrations for mice.
  • NOURO is a dietary supplement. It is not a treatment, cure or preventive for Alzheimer's disease or any other condition, and nothing on this page should be read as a claim that it is.
